DNA Barcoding with ONT MinION Course

This course will cover all the basics of DNA barcoding fungal specimens with the Oxford Nanopore Technologies (ONT) MinION. Topics included will be specimen collection and drying, taking tissue, DNA extraction, DNA amplification, DNA sequencing, and data analysis. It will cover all of the benefits, challenges, and limitations of the MinION system. No previous experience is required for this course. We will start from the basics. This will be a 1.5 day course, starting at 9am and continuing into the early evening on Saturday, with a half day on Sunday. This course will be held online. Every topic will be taught from the beginner perspective. It will also be taught from a practical perspective rather than a theoretical one. When baking a cake, you can follow a recipe and get to the desired end result. You do not need to fully understand all of the chemical reactions going on within the bread and how yeast ferments sugar to be successful baking. Think of this course in those terms. It will focus primarily on the practical lab aspects rather than the theoretical underpinnings (but there will be a bit of both).

All registration fees for this course will go to the Mycota Lab / Hoosier Mushroom Society’s efforts to barcode all of the fungi from North America in 2024. There will be a tiered admission for registration. Each tier has a limited number of registrants available.

After the course, each registrant in the USA will be offered a dual-indexed primer plate for their initial DNA barcoding efforts and to test their basic PCR skills. You would do the DNA extraction & amplification and send it back for sequencing. This plate will allow you DNA results for 96 specimens – which is typically more than the cost of the course. (You will need a thermocycler that accepts fully skirted PCR plates). This can be done at any time after completion of the course.

The goals of hosting this course are four fold. 1.) To give participants a working-level understanding of the DNA barcoding process with ONT MinION. 2.) To build the community of people that are proficient in working with nanopore sequencing. 3.) To help fund our continuing DNA barcoding efforts for North American macrogfungi. 4.) To fund future research and equipment that will allow continuous improvement and scaling of this project.
